发酵乳杆菌LF-8001高密度细胞培养的研究

摘要:试验利用从自然发酵泡菜中筛选出的一株性能优良的优势菌株乳杆菌LF-8001进行高密度培养,通过单因素试验和正交实验分析初始pH、接种量、培养温度、外加营养因子种类以及中和剂种类对乳杆菌LF-8001高密度细胞培养的影响。确定乳杆菌LF-8001的最佳高密度细胞培养工艺为:以2%的西葫芦汁液作为外加营养因子、0.5% K2HPO4溶液为中和剂,调节MRS培养基初始pH7,接种6%的乳杆菌LF-8001后于30 ℃恒温培养至稳定期,在此条件下乳杆菌LF-8001的菌浓度达到了1.08×1010 CFU/mL

AbstractA superior strain of Lactobacillus LF-8001 was selected from the natural fermented pickles for high cell density cultivation. The effects of initial pH, inoculum volume, culture temperature, kinds of additional nutritional factors and neutralizers on LF-8001 high cell density cultivation were analyzed by single factor and orthogonal experiments. The optimal high cell density cultivation technology of LF-8001 was determined as follows: 2% zucchini juice was used as an additional nutrient factor, 0.5% K2HPO4 solution was used as neutralizing agent, the initial pH of MRS medium was adjusted to 7, and 6% LF-8001 was inoculated, and then cultured to stable phase at 30 ℃. Under these conditions, the bacterial concentration of LF-8001 reached 1.08×1010 CFU/mL.
